Simulating rare events in systems without detailed balance

Abstract: Processes that happen rapidly, yet infrequently, are very difficult to simulate, because of the long waiting times between events. Although various methods have been developed for simulating rare events, these are generally only applicable to equilibrium systems where detailed balance is satisfied. Motivated by the problem of a bistable genetic switch, we have developed a method for calculating rate constants and obtaining transition paths in equilibrium or non-equilibrium systems. This "Forward Flux Sampling" method has been applied to the flipping of a genetic switch and the translocation of a polymer through a pore.
